Anti-complement activity is observed when patient serum fixes or destroys complement in the absence of any added test antigen. Such activity leads to inconclusive results in this complement fixation assay. Anti-complement activity may be due to a number of factors such as circulating immune complexes, immunoglobulin aggregation, rheumatoid factors, hemolysis, excessive freezing and thawing of the sample, detergent or microbial contamination in the specimen, and some medications.
